You have to unscrew the bottom rail, this is as difficult as you think, the screws will be rusted and hard to shift also possibly hard to find.
I have done this on several cars now and it takes time and patience. here is a good tip i found on the internet:
Sliding Window Channels.....
Sooner or later all you owners of Minis with sliding door windows are going to have to replace the black double channel that the glass slides in.
Those who have done it before will appreciate how difficult it can be to remove the screws holding the lower channel in place. The heads have usually rusted away, and the glass prevents easy access.
I’m afraid that you are still going to have to get those screws out, but, if you follow this tip, it should be the last time.
Basically, dismantle as usual, clean everything up, and re-assemble, but leave the double channel out! You will need to use screws with as flat a head as possible for this technique to work. Then, with the glass pushed near the back, feed the new flexy channel in from the front. It should slide under the glass without too much trouble. The glass will make sure that it doesn’t need fixing down.
Anytime that you wish to replace it in the future, just open the windows, prise up the front edge of the channel, and haul it out. Slide in the new one and the job’s done! No excuse for having moss growing in your window channels now!
Please note that this only works if you use the flexible type of window channel (as supplied by DSN Classics). It obviously won’t work with the rigid type.
Part numbers for the new channels are DRS3052H for the lower and DRS3053H for the upper. There is an alternative flocked rubber upper channel used on later cars. This is our part number DRS3066.

A good tip when reinstalling your sliding windows ....once you've repaired or sprayed the door spray waxoil in the gutter and make sure drain holes are clear ....
(1) get a tube of black sicaflex bonding compound and run a bead under new window channel stick this to new chrome catch runner
(2) mask off top outside front edge of door and inside top edge and run a bead of sicaflex along outter edge (door skin) and along inner edge .....
(3) now with the aid of a friend pop lager glass into middle of front channel of runner and small glass in inner channel now lift chrome channel and glass and with your friend help offer glass into top channels... now this is were you have to be carefull and lift chrome channel over beads (could be messy) and push right into place and clean off excess sicaflex and leave to cure...............good luck............
