If you have access to a vice clamp the trumpet in the vice being careful not to damage the soft alloy, give the knuckle joint a good soaking with penetrating fluid and using a cold chisel and hammer on the knuckle, as it will not matter if you damage the knuckle as I would hope you are replacing them, hitting the chisel, probably quite harm will eventually release the knuckle, may help if you keep rotating the trumpet so that the knuckle as it starts to loosen and come out will come out square.

A long drift through the middle, the bore for the knuckle is open ended inside the trumpet.

They can be stubborn to the point that almost no abuse will shift them (well not without damage to the trumpet anyway). Use of a blow torch can help a lot.
