What's it for? Won't it get blocked by the spacer and gasket?

The manifolds are often drilled to suit a number of different carbs. This manifold can use twin 1.5" (HS4 HIF38), 1.75" (HS6 HIF44), or H4

The H4 carbs are 1 1/2" and have the two vertical studs

Sorry I guess I wasn't clear. The hole is in the flange on the carb and you can see what looks like a vertical half pipe of brass , about 1/2" down the hole. At the bottom of the carb bore is a tiny hole below wear the butterfly stops at idle, with a brass tube liner that doesn't quite line up with the half pipe in the flange face hole.

They usually have an ally bung in the hole near the face, and as you say just get blanked off with the gasket / spacer anyway
