How do you know you have a misfire on cylinder 2?

My mini has a misfire on cylinder 2 when I remove plug cap and arc to plug it fires. I've replaced plugs points leads cap anyone have any idea?Thanks.
Not quite sure what you mean when you say when you arc the spark to the plug it fires. Do you mean the engine then runs on 4 cylinders, or do you mean you can hear the spark jump to the plug but the engine still doesn't run on 4?

It’s a little trick to test the system. If you pull the lead off and hold it off the top of the spark plug it’ll make a wider gap, which can increase the spark power and make the plug fire.
