If you're sure the screw is turning and still no change then you may have a totally inaccurate needle in the carb or needle excessively worn. Also I'd recommend removing then whipping the bottom off the Carb, (4 screws). At the bench with the carb off you should see the jet assembly moving up and down as you turn the mixture screw in/out, if there is no movement then you have a mechanical problem in the carb.
Do you know what Carb needle is in the hif44?

It could be oil that got into the bores/exhaust when you took the head off, it may need a good running session to burn the oil off.

If you can turn the screw on a HIF type SU with no difference in the CO reading then the lever has broken off inside the float chamber that raises and lowers the jet (common)

If you have a co meter you can see the mixture alter, otherwise by winding it out the engine will run weak and stutter/stall, or take the dashpot off and see if you can see the jet moving up/down.

If you have the carb setup and the timing is correct and its burning oil then points to all is well engine wise, have you pulled the plugs and had a look to see if they showing signs of excess fueling which could also make it smoke
